Newbie with spyware problem

I have had spyware problems in the past but never as bad as I am having now. I am a dork when it comes to this stuff so please explain a solution as if you were talking to a 5 year old. My problem is some sort of spyware is sucking my hard drive space at rapid paces. I thought it may be a virus so I ran a 17 hour (ugh!) virus scan (with current definitions) and came up clean. I ran spybot and geot rid of everything (including a DSO exploit) but when I boot back up the DSO exploit is back as is many other spyware crap. I have beenb able to successfully eliminate spyware in the past. I am not sure what woulds suck my hard drvie away (I lost 4.70 gigs in about 5 5 minutes!) and how to get rid of this junk! I have Windows XP and Nortons 2000 with the current releasew of spyware.
