Is there any way to disable files that are loaded in memory and locked by the operating system?

Can you give us an example of what files you're talking about?

Try rebooting in Safe Mode.
What sort of files are you tring to kill? If you have a virus/spyware problem there are other things you will need to do to stop it once and for all.

It was whilst using the program "eraser". A load of errors came up. Seems it couldent wipe the cluster tips because these files were locked.
