Boottime defrag on SATA

How do you get Diskkeeper to do a boottime defrag on a SATA hard drive? When I do that, it says that it cannot proceed because another process may be using it or it isn't NTFS.

However, I can do a boottime CHKDSK scan with Diskkeeper

This is on my new computer:
EPoX EP-MF570SLI
AMD Athlon 64 X2 3800+
Patriot Extreme Performance Low Latency DDR2 800 2GB (need to figure out how to get the RAM to run at 4-4-4-12, currently at 5-5-5-16)
Seagate 250GB SATA II
