You can if there is a source block down there. And from what I have seen it will still cause a flowing animation at the top of that column of blocks.
sounds like 3 high 1 out would work. Would take some testing. I'll hop on to re-optimize the spider grinder tonight and note behaviors while I'm at it.
Also if you pull water blocks from below the surface, you can create an underwater flowing effect, which can make it very difficult to swim.
